Orientation-specific Conjugation
This completed NTNU pilot project developed a novel strategy for producing protein A-based magnetic nanobeads with controlled, oriented protein attachment, addressing a key limitation of commercial antibody-purification beads, where unoriented binding often leads to batch variability and reduced performance.
Using a site-specific bioconjugation approach, the project engineered a modified antibody-binding protein domain and coupled it to functionalized magnetic nanoparticles in a defined orientation. Initial results showed promising improvements in antibody-capture efficiency compared to a leading commercial product, providing early proof-of-concept for a more effective and consistent alternative for antibody purification and related applications — a market valued in the hundreds of millions of dollars.
These findings laid the groundwork for the subsequent main project, which further developed and scaled the technology toward commercialization.
